Alas-7 ng umaga nagsimula ang operasyon sa dalawang pick up points, ang Parañaque Integrated Terminal Exchange (PITX) at ang BFCT East Metro Transport Terminal sa Marikina City.
Tatlo ang ruta na dadaanan ng mga bus, ang una na mula sa PITX ay maghahatid sa Makati Medical Center, St. Lukes BGC, Rizal Medical Center, The Medical City-Ortigas, East Avenue Medical Center, Philippine Heart Center, Lung Center of the Philippines at Quezon City General Hospital.
Ang pangalawang ruta ay mula PITX patungo ng Pasay City General, Adventist Medical Center Manila, Philippine General Hospital, Jose Reyes Memorial, Manila Doctors Hospital, UST Hospital, San Lazaro Hospital, Jose Reyes Memorial, at Chinese General Hospital.
Samantala, ang ikatlong ruta ay mula sa BFCT patungo ng Quirino Memorial Medical Center, East Avenue Medical Center, Philippine Heart Center, Lung Center, Providence Hospital, Capitol Medical Center, St. Lukes East Avenue, UST Hospital at San Lazaro Hospital.
Una rito, nagdeploy na rin ng mga sasakyan ang Philippine Coast Guard para maghatid ng mga health workers sa Maynila.
Samantala, nagset up na rin ng operations center ang DOTr para sa motor pool na gagamitin ng mga miyembro ng Inter-Agency Task Force (IATF) for the Management of Emerging Infectious Diseases.
